Effects of Five Roots Extract Against on Cisplatin-Induced Testicular Injury, Oxidative Stress in Rats
Abstract
: Population are getting older in the statistics by the WHO. Today for the first time in history most people can expect to live into their 60s and beyond. One of the main concerns of medical professionals and researchers is definitely healthy aging. The healthy aging is a key to longevity. The people of the Asia have been used various plant, aninal and mineral-originated medicines for rejuvenating and improving strength. There are about 300 formulas based on 99 different materials derived from plants, animals and minerals. The five roots have been used as rejuvenating remedies in traditional medicine. To examine the effects of aqueous and ethanol extracts of the five roots on cisplatin-induced testicular injury in rats. Aqueous (994 mg/kg) and 70% ethanol (874.4 mg/kg) extracts were given orally for 10 consecutive days. Cisplatin was administered intraperitoneally (7.5mg/kg) on the last day of the five roots treatment. Dehydroepiandrosterone (DHEA), Cinaspol were used as comparisons. Blood levels of testosterone were determined by enzyme-linked immunosorbent assay. Sperm motility and concentration were determined by Ds microscope and NIS-Elements. Caspase-3 and ВcL2 gene expressions were examined by Q-PCR. Testicular weights, sperm motility, sperm concentration and plasma levels of testosterone were significantly decreased in rats administered cisplatin (p<0.05) and these were significantly increased by aqueous and ethanol extracts of the five roots, DHEA, Cinaspol compared to untreated model. Aqueous and ethanol extracts of the five roots restore testicular functions by increasing blood levels of testosterone, sperm motility, sperm concentration, reducing testicular necrosis in rats administered cisplatin
